Common Issues when Using 32 Karat Software for Peak Integration

Answer

To improve peak integration with the 32 Karat software, please try the following suggestions:

1. For differences between integration events (used with the method file only or applied to multiple different data files in a reprocessing sequence) and manual fixes to individual data files:
      a. Please remember, fixes are per data file. Hence be sure that when fixes are applied, they only will apply to one data file.
      b. For multiple data files, try to determine similarities in data files and then add them to the integration events table only (and not additionally to the fix table).

2. Don't use multiple manual peak lines for fixes. Work the values for the integration events better to allow use of the method integration for the issue. 

3. Turn off the Caesar integration if your baseline looks fairly flat overall. To do this, first close the instrument control window (where Direct Control is). Then navigate to the icon that is normally double-clicked from the first window and right click. Select Configure->Instrument->Configure, and then click Options. Un-check the Caesar checkbox, and click OK. Open the instrument window again and see if there is an improvement after re-integration. The Caesar integration filter is a noise filtering algorithm and makes use of a centralized averaged baseline for all peaks, which does not work well for samples that already are fairly noise-free. 

4. Zoom in on the baseline as much as possible when assigning manual peaks. This allows for better peak start and stop times to show that the slope change comes from that start point and ends flat with the stop point.
