Updating Quantitation Method Files in Analyst® and Multiquant™ Software with a New Analyte

Answer

When adding new compounds to a MultiQuant™ or Analyst® software quantitation method, the other associated method files also need to be updated. Updating your quantitation method means, three other methods actually need to be updated. 

1) Update the Analyst acquisition method (.dam): Open the acquisition method that needs to be updated. Add the Q1 and Q3, RT and any other information needed in the table of transitions in the acquisition method for the new compound. Save the .dam file under a new name.

2) Update the Analyst batch templates (.qmf): New compounds added to the method will not update until the Quantitation field in the Batch Editor is set to none, and Quick Quant is selected. Name and save the batch file. The new drugs are now added to the .qmf file, but the drug concentrations will be empty.  The compound concentrations can be re-imported by copying the values from an Excel file with a simple [Ctrl]C and [Ctrl]V for each column.

3) Update the MultiQuant method (.qmethod): A new Multiquant method is created by acquiring samples with the new compounds, and then creating a new method based on the new data files. The Multiquant method can also be updated by exporting your old method to Excel and adding the new groups and internal standards and then importing this back into Multiquant. You will need to have the first internal standard chosen in the drop down menu to be able to copy and paste the entire table from Excel.

Once the new .dam, .qmf, and .qmethod files are created, save the batch template with the new acquisition method and new quantitation method so that they are always used in future work.
